  • Generating biomolecular mixtures at specified mixing ratios is an essential part in developing high-throughput microfluidic live cell screening platforms. However, producing three or more reagent mixtures require complicated structures and operating procedures when using traditional microfluidic network-based gradient generators. Here, we present an on-demand geometric metering-based mixture generator which facilitates robust, scalable, and accurate multi-reagent mixing in a highthroughput fashion, where mixing ratios are intrinsically determined by the volume ratios of corresponding reagent chambers. A high-throughput live cell array combining this mixture generator with single-cell trapping arrays was implemented for studying combination effects of environmental toxins with single-cell resolution analysis.
