abstract

  • We have developed a multi-channel in vitro blood-brain barrier (BBB) platform that can better mimic the in vivo physiological conditions and responses. To decipher signals involved in cell to cell interactions, the compartmentalized multi-culture microsystem is developed to have the capability to accurately control the positioning of multiple cell types and to be able to form high-quality tight junctions. The improved brain-on-a-chip is enabled by utilizing in vivo level of blood flow shear stress values comparable to the condition experienced by an in vivo BBB structure, as well as through the use of primary mouse endothelial cells with primary astrocytes. The multi-channel structure also makes this chip amenable to higher throughput studies.
