A method is described by which radioactivity in proline-C14 and hydroxyproline-C14 derived from biological materials may be determined simultaneously. The method involves oxidizing both imino acids with chloramine-T and then selectively extracting the oxidation products into toluene. Of the amino acids derived from proline in biological systems, glutamic acid does not interfere at all while ornithine gives rise to an oxidized product which is toluene extractable to some extent. The method is extremely useful in studying the metabolism of hydroxyproline-containing proteins, where interference from ornithine is of no major importance. 1962.
