Statement of Purpose: Protein and DNA have the potential to self-associate in pre-defined patterns, creating ordered composite materials with multiple avenues for functionalization. Such materials require a protein component that can both self-assemble and bind specific DNA sequences with high affinity. Drosophila Ultrabithorax (Ubx) is a transcription factor which self-assembles into a variety of materials in vitro. Here we test if biomaterials composed of Ubx are capable of reversible DNA binding via the Ubx homeodomain, co-assembly with DNA into materials, extended release of DNA, and delivery of DNA to cells.
