  • The objective of this study was to prepare, characterize and perform in-vitro release studies of indomethacin micromatrices with mixture of Eudragits, RS100 and L100. The procedure involved dissolution of drug and polymer mixtures in alcohol USP followed by the addition of water (reduced to pH 1.2, at 4C) at controlled rates. The mixture was stirred to obtain micromatrices which were filtered and air-dried. The micromatrices were then compressed to obtain biconvex tablets with a diameter of 7 mm. A seven-factor 12-run Plackett-Burman screening technique was employed to evaluate the main effects of rate of addition of water, drug to polymer ratio, polymer to polymer ratio, alcohol to water ratio, amount of filler added, stirring speed (rpm) and compression pressure. Response variable studied was dissolution T50 values. The factors with maximum influence on the response in decreasing order are percent of additive, solvent ratio, drug to polymer ratio and compression pressure. The micromatrices were characterized by X-ray diffractometry and IR. The IR data did not indicate significant difference between the micromatrices and pure drug, but the X-ray diffractometry did. The drug showed reduced crystallinity in micromatrices. Results of the Plackett-Burman screening, X-ray diffraction and IR study are reported.
