  • 2016 Elsevier Inc. All rights reserved. Recent advances in single-particle electron cryo-microscopy (cryoEM) allow structure determination of macromolecular complexes to 3-4. resolution. In this article, we highlight the latest developments of hardware and software for single-particle cryoEM studies from nanometer to near-atomic resolutions. We describe some cautionary notes about hurdles often encountered in each step of the cryoEM experiment and data processing. We present examples of the macromolecular complexes studied by single-particle cryoEM to demonstrate the unprecedented structural resolution and the power of this method to determine structures in different biochemical states through a combination of biochemical and computational approaches.
