Metabolic Differences in Nonessential Amino Acid Plasma Concentrations in High- and Low-Active Mice Conference Paper uri icon

abstract

  • BACKGROUNDWith current health problems such as obesity and Type II Diabetes attributable to both physical inactivity and metabolic disorders, there continues to be a lack of information regarding physiological mechanistic interactions between physical activity and metabolism.PURPOSETo conduct basic metabolic analysis on mice either high active (HA) (C57L/J) or lowactive (LA) (C3H/HeJ), and determine basic metabolic differences between essential amino acids (EAA) in these activity types.METHODSAll procedures were approved by the TAMU IACUC. At 12wks of age, 20 HA male mice (body weight: 27.51.2g; lean mass: 22.51.3g; fat mass: 2.50.7g; food intake: 3.40.8g) and 23 LA male mice (body weight: 25.8 1.2g; lean mass: 21.01.1g; fat mass: 2.50.5g; food intake: 3.01.0g) (Jackson Labs) under anesthesia had a catheter placed in the right jugular vein for blood sampling. Plasma EAA concentrations were determined by LCMS/MS. Data were analyzed using unpaired ttests for each EAA.RESULTSLA mice had significantly higher EAA for THR, VAL, MET, ILE, LEU, TRP and HIS (Table). LYS was the only essential AA with a significantly lower concentration.CONCLUSIONWe hypothesize that the consistent higher EAA plasma concentrations in LA strain mice indicate a modified essential AA metabolism that is linked to their lower activity levels.Support or Funding InformationStudy funded by research development grant from the VicePresident of Research, Texas A&M University Amino Acid Low Active (n=20) High Active (n=23) Difference P Threonine (THR) 206.7 9.8 161.4 6.7 45.3 11.9 0.0005 Valine (VAL) 187.9 7.1 156.8 4.9 31.1 8.6 0.0008 Methionine (MET) 60.9 2.7 47.3 1.5 13.6 3.1 0.0001 Isoleucine (ILE) 57.7 2.3 48.5 1.9 9.2 3.0 0.0041 Leucine (LEU) 144.8 6.4 125.2 4.8 19.6 8.0 0.0184 Tryptophan (TRP) 71.1 3.0 58.2 2.3 13.0 3.8 0.0016 Phenylalanine (PHE) 86.7 4.3 77.5 2.4 9.2 4.9 0.0724 Histidine (HIS) 109.3 3.9 92.4 2.8 16.8 4.8 0.0011 Lysine (LYS) 379.9 17.2 481.4 18.2 101.5 25.0 0.0002 Sum EAA 1305.0 56.7 1248.7 45.5 56.3 73.1 0.9139 Plasma concentrations in m.
